Exploring the relationship between dynamics and stability of NIST monoclonal antibody 

Monoclonal antibodies (mAbs), are extremely attractive bio pharmaceutical products due to enhanced biocompatibility and target specificity. The ability of mAb products to maintain long term stability, or shelf life, is essential to ensure safety and efficacy can be maintained throughout all of the time required for shipping, handling, etc. Sufficient stability ensures that equitable distribution of these products is possible. Many studies have suggested that mAb stability is related with the dynamic fluctuations of each mAb. The goal of this research is to elucidate the nanoscale dynamics of mAbs in an effort to underpin the mechanism of stability and shelf life. This work will combine the power of neutron scattering techniques and the pre-competitive reference material, NISTmAb RM 8670. Findings, from this work conducted on NISTmAb, will be public access and serve to represent the majority of mAbs on the market today, (as an IgG1 mAb). This work is the first step toward the development of a rapid (days) predictive tool of therapeutic long-term stability (years). The overarching goal of this, would be to lower the cost and time involved in therapeutic development, to expand treatment access and increase number of treatments available.
